ABSTRACT

Objective:To investigate the dynamic changes of routine laboratory parameters during the course of hemorrhagic fever with renal syndrome (HFRS) and estimate the predictive value for the severity of the disease.Methods:A retrospective cohort study was conducted, which enrolled 394 HFRS patients admitted to the Second Affiliated Hospital of Air Force Medical University (374 cases) and the Second Affiliated Hospital of Xi′an Jiaotong University (20 cases) from January 2019 to January 2022. The patients were divided into mild (mild and moderate) and severe (severe and critical) groups.The basic information, personal history, past history, treatment, complications and other clinical data of patients were collected and the results of the laboratory examinations in the morning at day 1, 2, 3, 4, 5, 7, 10, 15, 20 and 25 of hospitalization and before discharge were recorded. The dynamic changes of the patients′ routine laboratory indicators and the dynamic predictive values of each indicator for severe condition were analyzed. Mann-Whitney U test and chi-square test were used for comparison, and receiver operator characteristic (ROC) curve was used for predictive value evaluation. Results:The age of 212 patients in the mild group was 38(27, 61) years, and that of 182 patients in the severe group was 49(32, 64) years, the difference was statistically significant ( Z=-2.24, P=0.025). The incidences of acute pancreatitis, acute respiratory distress syndrome, multiple organ dysfunction syndrome, the utilization rates of blood purification and mechanical ventilation in the severe group were 6.0%(11/182), 12.6%(23/182), 19.8%(36/182), 89.6%(163/182) and 22.5%(41/182), respectively, and those in the mild group were 0(0/212), 0(0/212), 0(0/212), 15.6%(33/212) and 0.5%(1/212) respectively, and the differences were all statistically significant ( χ2=13.18, 28.45, 46.15, 214.48 and 50.02, respectively, all P<0.05). The levels of white blood cell count, lymphocyte count, monocyte count and neutrophil count were all increased rapidly after onset and peaked at days 4 to 6 of illness, with the counts of 14.2(9.7, 20.7)×10 9/L, 4.2(2.3, 6.2)×10 9/L, 1.5 (0.8, 3.3)×10 9/L and 8.3(4.3, 11.4)×10 9/L, respectively. Aspartate aminotransferase peaked (102(66, 178) U/L) within three days after onset and then decreased rapidly, returned to normal level by day 12. Blood urea nitrogen and creatinine both increased steadily after onset, peaked at day 9 to 10, with the levels of 13.2(7.7, 19.1) mmol/L and 255.4(122.9, 400.9) μmol/L, respectively. Prothrombin time, activated partial thromboplastin time, fibrinogen degradation products and D-dimer levels at day 3 after onset were 12.7(12.0, 13.2) s, 38.7(33.5, 51.9) s, 12.6(6.9, 32.0) mg/L and 4.9(2.2, 13.7) mg/L, respectively.Platelet count at day 4, neutrophil count at day 5, creatinine at day 11 and blood urea nitrogen at day 14 after onset had decent predictive values for estimating severity, of which the area under curve (AUC) values were 0.801(95% confidence interval (95% CI) 0.727 to 0.875), 0.824(95% CI 0.770 to 0.878), 0.862(95% CI 0.805 to 0.919) and 0.810(95% CI 0.722 to 0.897), respectively. Conclusions:Routine blood count, liver function and coagulation are important reference indicators for early warning of severe disease of HFRS, while with the progress of the disease, renal function indicators are effective in differentiating the severity of the disease. The platelet count at day 4, neutrophil count at day 5, creatinine at day 11 and blood urea nitrogen at day 14 after onset have predictive values for severe HFRS.

ABSTRACT

Left ventricular assist device(LVAD) can improve the survival rate and quality of life of patients with end-stage heart failure, but the quality of life of patients after LVAD implantation is still lower than that of heart transplantation. The reasons for the influence of LVAD implantation on patients' quality of life include discomfort symptoms, surgical complications and high re-hospitalization rate, limitation of physical activity level and negative psychological emotions after device implantation. Cardiac rehabilitation training can improve the quality of life of LVAD patients by improving their psychological state, enhancing their exercise ability and reducing the risk of re-hospitalization. The exercise prescription of LVAD implant patients can include aerobic endurance training, walking training, light to moderate resistance training, gymnastics training, etc. The exercise training intensity should be controlled at the first ventilation threshold(anaerobic threshold), 50% peak oxygen consumption or 12-14 points of Borg scale. In addition, the safety of cardiac rehabilitation training for LVAD patients has been fully demonstrated.

ABSTRACT

With the range of application of computational biology and systems biology gradually expanding, the complexity of the bioprocess models is also increased. To address this difficult problem, it is required to introduce positive alternative analysis method to cope with it. Taking the dynamic model of the epidemic control process as research object, we established an evaluation model in our laboratory. Firstly, the model was solved with nonlinear programming method. The results were shown to be good. Based on biochemical systems theory, the ODE dynamic model was transformed into S-system. The eigen values of the model showed that the system was stable and contained oscillation phenomenon. Next the sensitivities of rate constant and logarithmic gains of the three key parameters were analyzed, as well as the robust of the system. The result indicated that the biochemical systems theory could be applied in different fields more widely.

ABSTRACT

Objective To evaluate bacteriophage therapy for gut-derived sepsis caused by Acinetobacter baumannii in mice.Methods Lyric phages of Acinetobacter baumannii were isolated from environment by using double-layer agar method.A murine model of gut-derived sepsis was established by oral administration of ampicillin-resistant Acinetobacter baumannii and injection of ampicillin and cyclophosphamide into peritoneal cavities of mice.Bacteriophage therapy were given 1 d before infection (group 1),2 d(group 2) and 6 d(group 3) after infection.The survival of the mice was observed,mice without bacteriophage therapy were as control.Independent-sample t test was performed to compare inflammatory cytokines levels in peripheral blood and liver,number of bacteria in liver and spleen between mice with and without bacteriophage therapy.Results The minimal lethal dose of Acinetobacter baumannii for mice with gut-derived sepsis was 1 × 107 CFU/mL.The survival of the mice in group 2 (4/6 survived),which were treated with bacteriophage 2 d after inoculation of Acinetobacter baumannii,was higher than those of group 1 (2/6 survived),group 3 (3/6 survived) and the control group (phage-untreated,0/6 survived).Interleukin (IL)-1 β,tumor necrosis factor (TNF)-α and IL-6 in peripheral blood in mice with bacteriophage therapy were (105 ±6) ng/L,(105 ± 11) ng/L and (104 ± 12)ng/L,which were lower than those in control group (t =5.04,9.05 and 9.33,P < 0.01) ; IL-1 β and TNF-α in liver of mice with bacteriophage therapy were (104 ± 9) ng/L and (104 ± 11) ng/L,which were lower than those in the controls (t =13.70 and 12.80,P <0.01),but IL-6 levels were not of statistical difference between therapy and control groups (t =1.06,P > 0.05).Number of bacteria in liver and spleen in mice with bacteriophage therapy were (2.9 ± 1.3) × 103CFU/g and (8.3 ±7.6) × 102 CFU/g,which were also lower than those in control group (t =9.16 and 8.96,P < 0.01).Conclusions Bacteriophage therapy can be effective against gut-derived sepsis caused by Acinetobacter baumannii.

ABSTRACT

Objective To determine the effects of sexual hormone therapy at varied doses on prevention of bone mineral loss in Chinese postmenopausal women. Methods From March 2002 to March 2003, 90 Chinese postmenopausal women were randomly divided into three groups, each given one of the following regiments for 12 months, estradiol valerate (EV) 1 mg plus medroxyprogesterone (MPA) 2 mg for group A (31 subjects), eonjngated estradiol (ethinylestradiol-3-cyclopentylether, CEE) 0.45 mg plus MPA 2 mg for group B (29 subjects) and livial 1.25 mg for group C (30 subjects), respectively. In addition, 400 mg of elemental calcium were given daily to all those women. Bone mineral density (BMD) of the 2nd to 4th lumbar vertebra (L2~4) and biochemical markers of bone turnover, urine N-telopeptide of type Ⅰ collagen/creatinine(NTX/Cr) and serum total alkaline phnsphatase (ALP), were measured before and after drug administration. Results After treatment for 12 months, BMD of the L2~4 increased significantly by 0.039 g/cm2(P<0.01) in group B, but not significantly in group A or group C (P<0.05). Increases in BMD of the L2~4 was more in group B than that in group A and group C, respectively (P < 0.05), but no significant difference in BMD increase between group A and group C was found (P >0.05). After treatment for 6 months, urine NTX/Cr reduced from the baseline for all the three groups (P < 0.05), but no significant difference among group A, group B and group C was found (P >0.05). After treatment for 12 months, serum ALP significantly reduced from the baseline for all the three groups (P <0.01), but no significant difference among group A, group B and group C wag found (P > 0.05). Conclusions Sexual hormone therapy at varied doses lower than regular one for 12 months was effective in preventing bone mineral less in postmenopausal women.

ABSTRACT

OBJECTIVE: To establish the HPLC method for the content determination of paenol in Liuwei dihuang soft extract. METHODS: The octadecylsilyl silicagel was used as filter. Mobile phase consisted of methanol-water(70 ∶ 30) at detection wavelength of 274 nm. The flow rate was 0.8 mL?min-1 and the column temperature was room temperature. RESULTS: The linear range of paeonol was 50~300 ?g?mL-1(?g?mL-1) with an average recovery of 99.63%(RSD=1.26%, n=6). CONCLUSION: This method is sensitive, accurate and reproducible for quality control of Liuwei dihuang soft extract.

ABSTRACT

Objective To establish stable insect cell line expressing mIL4 on the basis of construction of inset expression vector pIZT/V5-His.Methods Amplified mIL-4 and pIZT/V5-His were treated with EcoRI and XbaI,and mIL-4 were ligated into pIZT/V5-His vector using T4 DNA ligase.Sf9 cells were transfected with recombinant DNA and ELISA was employed to detect soluble mIL-4 production by transfected Sf9 cells.Results Transfected Sf9 cells could significantly produce mIL4(1 mg?L~(-1)) compared!with control group(0.003 mg?L~(-1)).Conclusion Inset expression vector pIZT/V5-His is an ideal expression vector for mIL-4 production in transfected cells.

ABSTRACT

Objective To construct the double hairpin siRNA and green fluorescent protein(GFP) expressing vector pU6/double-siRNA/Neo/GFP/1A/2A to interfere 1A and 2A gene of Coxsackie virus B4.Methods 21 bp fragments of the Coxsackie virus B4 2A and 1A gene were chosen as the targets and 65 bp complimentary fragments were synthesized,then the target gene fragments were cloned into pSilencer2.1U6 Neo and pGCsi-U6/Neo/GFP/siNeGative,respectively,then the double siRNA expressing vector pU6/double-siRNA/Neo/GFP/1A /2A was constructed by restrict endonuclease digestion,elctrophoresis isolation and reclaimer,ligatied by T4 DNA ligase;then the double siRNA expressing plasmid was transfected into Hela cells,and the GFP was observed under fluorescent microscope.Results The correct results showed that the recombinant plasmid had the correct special fragments and DNA sequence detected by restrict endonuclease digestion, electrophoresis and DNA sequencing;and GFP was also observed in Hela cells tansfected with pU6/double-siRNA/Neo/GFP/1A /2A under fluorescent microscope more than 15 d after transfection.Conclusion The double siRNA expressing vector pU6/double-siRNA/Neo/GFP/1A/2A is constructed successfully;it has the correct target viral gene sequences and can express GFP gene in Hela cells more than 15 d after transfection.

ABSTRACT

Objective To explore the influence of different domains of STAT6 expressed by the vector pGEX-6p-2 on signal transduction.Methods Different domains of STAT6 were amplified by PCR technique and ligated respectively into the prokaryotic expression vector.Recombinant DNA was transformed into HB101 cells.Transformants were lysed by supersonic method and lysates run on SDS-PAGE.Results Western blotting analysis showed a high level expression(60 000,56 000 and 50 000) of different domains of STAT6 SHST,DST and NST,and the empty vector only had GST expression.Conclusion Expression vector pGEX-6p-2 can be used for high level expression of different domains of STAT6.

ABSTRACT

Objective:To determine distribution of fungal florae of candial balanoposthitis (CBP),the relatioship between antibiotics administration and candial infection and the major source of opportunistic pathogens.Methods:The fungal strains were collected and identified,and the history of antibiotic administration was reviewed.Results:①Foreign fungi were predominant pathogens of CBP;②Dysbacteriosis induced by antibiotics stimulated the growth of foreign fungi,which resulted in CBP;③There was a positive relationship between fungal distribution of male patients and their sexual partners.Conclusion:In order to reduce the incidence of CBP safe coition and reasonalbe administration of antibiotics should be emphasized.
